    Fabsorbent™ F1P HF is a mixed-mode chromatography resin, that provides an alternative purification solution for the effective capture of differing antibody binding regions. Unlike expensive protein A or protein L strategies, Fabsorbent™ F1P HF uses a synthetic ligand that offers a cost-effective alternative for antibody fragment and bispecific separation.

    Fragmented antibodies are smaller antibody fragments that consist of portions of the heavy and light chains, specifically one constant and one variable domain from each. Unlike full-length monoclonal antibodies (mAbs), these fragments lack the Fc (fragment crystallizable) region, which significantly reduces their size. This lack of the Fc region enhances their ability to penetrate tissues more effectively, making them especially valuable for therapeutic applications where rapid distribution and clearance are important.

    The absence of an Fc-region in antibody fragments complicates their purification, where traditional methods like Protein A chromatography used for full antibodies, are not applicable.     The recommended storage condition is 2 – 30°C in 20% ethanol.

    How is Fabsorbent™ F1P HF regenerated?
    Removal of any residual adsorbed material can be achieved by washing the column with 1.0 M NaOH. A contact time of 1 hour will normally suffice to ensure destruction of viable organisms, although up to 5 hours contact time may be required. No less than 5 column volumes are recommended. Once cleaning with NaOH is complete, wash with at least 3 bed volumes of equilibration buffer until the pH and conductivity of the column eluate is equal to that of the buffer entering the column. Complete this process prior to further use or storage in the storage buffer.
