Antibodies

Antibody therapeutics have become a cornerstone of modern medicine, used to treat everything from cancer to autoimmune and infectious diseases. Monoclonal antibodies (mAbs) remain the most widely used format, supported by decades of development and well-established manufacturing platforms. Yet even with their success, mAbs still present unsolved purification challenges—from handling host cell process related-impurities, aggregation, and the variability between differing expression systems.  

As antibody engineering evolves, new formats like antibody fragments (Fab, scFv) and bispecific antibodies (bsAbs) are expanding what’s possible. These formats enable better tissue penetration, faster clearance, or simultaneous targeting of multiple disease mechanisms. However, their structural differences introduce complexity in downstream processing. Traditional affinity methods like Protein A aren’t always applicable, and new formats often require customized approaches to ensure purity, yield, and scalability.  

A flexible, toolbox-based purification strategy is key to unlocking the full potential of mAb and mAb-derived therapeutics.

    Fabsorbent™ F1P HF

    Fabsorbent™ F1P HF is an appealing, mixed-mode alternative to Protein L for purifying antibody light chain fragments, such as kappa and lambda light chains, Fab and scFv fragments. Using a unique synthetic ligand that demonstrates broad specificity for capture and purification of mAbs and their derivatives, Fabsorbent™ F1P HF has an affinity for both kappa and lambda chains. The synthetic nature allows for robust base-stability, and the PuraBead® 6HF base matrix offers stable flow properties even for large-scale manufacturing. These attributes make it an appealing alternative to expensive Protein A and Protein L ligands as a platform technology.

    MAbsorbent® A2P HF

    MAbsorbent® A2P is a mixed-mode resin that gives an alternative purification approach in antibody purification by combining hydrophobic and hydrogen-bond interactions, often removing impurities such as antibody aggregates. It binds all four human IgG subclasses—including IgG3—which some other resins can miss. With high purity, strong viral clearance, and broad IgG compatibility, it’s a great fit when you need a more versatile approach to antibody workflows.

    Aminophenylboronate

    Aminophenylboronate is an alternative for antibody purification when you need to target glycosylated forms, such as IgG or remove glycosylated impurities. It is especially useful for capturing glycoprotein variants of antibodies or cleaning up unwanted glycated species from complex mixtures, like those expressed in yeast. The resin works by binding cis-diol-containing structures on antibodies and related impurities, allowing for selective capture or polishing. With a stable PuraBead® base, it delivers good flow and is easy to clean, making it a reliable option for refining antibody prep workflows.

      HCPure™

      HCPure™ is a mixed-mode resin designed with the removal of host cell proteins (HCP) in mind. This unique resin is composed of a unique mixed-mode ligand attached to PuraBead® 6HF agarose bead support. HCPure™ has a unique operational space compared to other commercially available products and can provide alternative strategies for the removal of problematic HCPs. Whilst selectively binding host cell proteins, HCPure™ can also remove host cell DNA, endotoxin, light chain IgG, and high molecular weight aggregates, to yield highly purified biological products with minimal target protein losses.  Due to its highly selective nature and ability to be tuned, by varying the chromatography conditions, HCPure™ removes impurities from a number of different expression systems, including, but not limited to CHO, E. coli, HEK 293, Pichia Pastoris, Saccharomyces Cerevisiae and Spodoptera frugiperda (Sf9 and Sf21).

      PuraBead® IEX

      Ion exchange (IEX) resins, such as Q PuraBead® or SP PuraBead® Edge, are a staple in antibody purification, often used after Protein A capture to remove impurities like host cell proteins (HCPs), DNA, and aggregates. They work by separating molecules based on charge differences, making them great for fine-tuning purity and removing closely related contaminants. But because antibody properties vary widely, there’s no one-size-fits-all solution—trying different resin types and formats is often key to finding the best fit for your process.

      PuraBead® HIC

      Hydrophobic interaction chromatography (HIC) resins help clean up antibodies by separating out aggregates, misfolded variants, or other hydrophobic impurities that are hard to remove with ion exchange alone. They work under mild conditions using salt gradients, which helps preserve antibody stability. Antibody properties vary widely, there’s no one-size-fits-all solution—trying different resin types and formats is often key to finding the best fit for your process.
