Antibodies

Today, monoclonal antibodies (mABs), antibody fragments, multi-specific antibodies, and antibody drug conjugates form the most significant category in the recombinant protein products sector, representing roughly half of the therapeutic proteins market. The downstream processing of these recombinant antibody products is vital, not only for achieving maximal yields and productivity but also for meeting the rigorous purity standards required for regulated therapeutic products. While Protein A affinity chromatography is a key component of many antibody purification processes, it is often insufficient on its own and inapplicable to a growing number of engineered antibodies that lack a Protein A binding site.

At Astrea Bioseparations, we provide a suite of chromatography adsorbents designed to work synergistically with Protein A, ensuring the attainment of the desired antibody purity at an affordable price. We work to craft solutions that address the unique challenges of antibodies that are not amenable to traditional Protein A chromatography.

Our Solutions

Capture and purify a diverse array of antibodies and antibody fragments with Fabsorbent™ F1P HF

Fabsorbent™ F1P HF is designed to bind antibodies containing kappa and/or lambda light chains, offering a versatile and convenient solution for the purification of various antibody fragments such as Fab, Fab', F(ab')2, scFv, and dAb antibodies. Built on the robust, alkali-stable PuraBead® technology, Fabsorbent™ F1P HF delivers superior performance compared to traditional Protein L affinity resins. One of its key advantages is the dual binding capability to both lambda and kappa light chains, making it an ideal choice for researchers and manufacturers seeking reliable, efficient, and broad-spectrum antibody purification solutions.

Capture and purification of human and mammalian IgGs with MAbsorbent® A2P HF

This robust, fully alkali-stable, PuraBead®-based affinity resin excels as either an initial IgG capture step from feedstocks free from Pluronic F68 surfactant or as a secondary purification/polishing step following Protein A or ion-exchange capture. The MAbsorbent® A2P ligand, engineered to mimic essential amino acid residues of Protein A, targets the same IgG region, achieving comparable purities and viral clearance, often with enhanced removal of antibody aggregates. Uniquely, it also binds all four subclasses of human IgG, including IgG3, offering a versatile and reliable solution for diverse bioprocessing needs.

Capture and separation of glycosylated antibodies with Aminophenylboronate

Aminophenylboronate is a fully alkali-stable resin featuring a meta-aminophenylboronate affinity ligand on a PuraBead® P6XL base matrix. This resin forms reversible complexes with sugars containing cis-diols, commonly found in glycan groups (including mannose, galactose, glucose, fucose, N-acetyl galactosamine, N-acetyl glucosamine, and sialic acid), making it exceptionally useful for selectively capturing glycosylated antibodies. It effectively separates glycosylated from non-glycosylated antibodies, or resolves glycosylated antibodies based on their sugar composition, thereby enhancing the specificity and efficacy of antibody-based therapies.

Removal of host cell proteins with HCPure™

HCPure™ utilizes a unique mixed-mode ligand from Astrea Bioseparations’ Chemical Combinatorial Library® and binds to proteins by a combination of hydrophobic and electrostatic interactions. HCPure™ targets a broad spectrum of host cell proteins from various cells (e.g. CHO, HEK293, E. coli), while minimally interacting with key biomolecules like antibodies and viral vectors. This fully alkali-stable resin supports high flow rates and offers extended operational lifetimes, making it an ideal choice for the polishing step following Protein A capture, ensuring high yields of exceptionally purified antibodies.

Polishing to remove residual impurities using PuraBead® IEX

Astrea Bioseparations’ Ion Exchange (IEX) chromatography is particularly well suited as a polishing step following Protein A capture, adeptly handling the capture and removal of product-related impurities and host cell proteins. We provide a comprehensive suite of robust, fully alkali-stable ion exchange resins crafted from our free-flowing, near-monodisperse PuraBead® 6HF base matrix. These resins are renowned for their high binding capacities, long operational lifetimes, high flow rates, and low back pressures. Simplifying the selection process, our pre-packed RoboColumns® and IEX Selection Kits make it easy to find the perfect PuraBead® ion exchange resin and chromatography buffers for your application, ensuring reproducible high yields of highly purified antibodies.

      Achieving clarity with PuraBead® HIC

Our Hydrophobic Interaction Chromatography (HIC) resins provide an effective polishing step post-Protein A capture, often used in tandem with ion exchange chromatography. These resins excel in the capture and removal of product-related impurities and host cell proteins, supported by our robust, fully alkali-stable, high-quality PuraBead® 6HF base matrix. These resins maintain high binding capacities and operational longevity while facilitating high flow rates and low back pressures. With our pre-packed RoboColumns® and HIC Selection Kits, selecting the ideal PuraBead® HIC resin and buffers is straightforward, consistently leading to the reproducible achievement of high yields of highly purified antibodies.
